Abstract
Diosmetin, is a bioactive flavonoid component of numerous traditional Chinese
medicinal herbs such as dandelion and
honeysuckle, which has clear
anti-inflammatory properties. Based on this efficacy, we examined the
anti-inflammatory potential of diosmetin using network pharmacology and in vitro
validation. Network pharmacology analysis screened out 41 high-probability
anti-inflammatory targets of diosmetin. Enrichment analyses showed that these
targets were mainly related to reactive nitrogen metabolism, and the NF-κB
cascade served as a dominant hub. In primary bovine mammary epithelial cells (bMECs)
model, it was confirmed that diosmetin pre-treatment suppressed IκBα and p65
phosphorylation, blocked NF-κB/iNOS/NO cascade and dimimished the production of
IL-1β, TNF-α and IL-6. Our results show that diosmetin exerts anti-inflammatory
effect against S. aureus infection in
bMECs by targeting theNF-κB/iNOS/NOsignaling axis, providing a new mechanistic basis for its potential in
clinical veterinary practice
