Brucellosis is a major zoonotic disease, causing significant losses in livestock
production, especially in low- and middle-income countries. In Pakistan,
epidemiological information on bovine brucellosis is scattered, preventing
effective disease control. In this study, the seroprevalence of bovine
brucellosis was determined, and the associated risk factors were identified in
Faisalabad Division, Punjab, Pakistan. The study was designed as a
cross-sectional study, with a sample size of 1,000 collected from four
districts: Faisalabad, Toba Tek Singh, Jhang, and Chiniot. Rose Bengal Plate
Test (RBPT), Serum Agglutination Test (SAT), and indirect ELISA (iELISA) were
used to detect anti-Brucella antibodies in serum. Epidemiological data
were collected and analyzed using bivariate analysis and multivariable analysis.
Seropositivity was 17.90% by RBPT, 16.30% by SAT and 14.70% by iELISA.
Inter-test agreement was highest between RBPT & SAT (95.8%), followed by SAT &
iELISA (93.4%) and RBPT & iELISA (92.4%). Multivariable analysis using
generalized linear mixed-effects models identified larger herd size, older age
(>8 years), male sex, pregnancy status, and repeat breeding history as
significant independent risk factors for seropositivity (P<0.05). These findings
confirm that bovine brucellosis is endemic in Faisalabad Division and highlight
the need for active surveillance, strategic vaccination, and improved farm
biosecurity.
